De Lamborghini Countach 5000 QV was de laatste grote technische stap in de ontwikkeling van de Countach voordat in 1988 de 25th Anniversary-versie verscheen. Lamborghini presenteerde de Quattrovalvole in 1985. De auto bouwde voort op de Countach 5000 S, maar kreeg een ingrijpend gewijzigde twaalfcilinder. De carrosserievorm bleef trouw aan het oorspronkelijke ontwerp van Marcello Gandini voor Bertone, al was de QV direct herkenbaar aan de hogere motorkap met de markante bolling boven het motorcompartiment. Lamborghini produceerde de Countach 5000 QV van 1985 tot 1988. Officiële merkgeschiedenis noemt 631 gebouwde exemplaren.
De techniek was de kern van de update. De V12 groeide naar 5,2 liter en kreeg, zoals de naam al aangeeft, vier kleppen per cilinder. Daarmee kwam het vermogen uit op 455 pk in de carburateurversie; voor de injectieversie werd 420 pk opgegeven. De motor lag nog steeds in lengterichting voor de achteras, met de versnellingsbak vóór het blok en een aandrijfas die door het carter naar het achterdifferentieel liep. Die lay-out was al sinds de eerste Countach kenmerkend voor Lamborghini en bleef ook bij de 5000 QV behouden.
De Quattrovalvole was niet alleen sneller dan zijn voorgangers, maar ook visueel nadrukkelijker. De bredere wielkasten, NACA-luchtinlaten en vaak gemonteerde achtervleugel sloten aan bij het straatbeeld van de tweede helft van de jaren tachtig. Tegelijk bracht de motorwijziging een praktisch nadeel mee: de naar beneden gerichte Weber-carburateurs vroegen extra ruimte, waardoor de zichtbaarheid naar achteren verder werd beperkt. Juist dat compromis typeert de auto. De Countach 5000 QV was niet ontworpen om alledaags te zijn, maar om maximale indruk te maken.
In historisch perspectief geldt de 5000 QV als de technisch rijpste Countach vóór de laatste facelift. Hij combineerde de oorspronkelijke Gandini-lijnen met de sterkste atmosferische V12 die de Countach tot dan toe had gekregen. Daarmee werd de Quattrovalvole het sluitstuk van een ontwikkeling die in 1971 begon als concept en uitgroeide tot een van de bepalende supercars van zijn tijd.
